Penambahan Nutrisi Pottato Dextrose Agar pada Pembuatan Starter Mikroorganisme Jamur dengan Bahan Baku Tepung Beras The Pottato Dextrose Agar Enrichment for Nutrients on Making Fungi Starter in Raw Materials of Rice Flour

Abstract
Rice flour is a raw material that is often used for starter. Rice flour has a starch content of 90% of dry weight. Starter is collection of mushroom cultur that is ready to be inoculatedfor fermentation. Mushrooms commonly used in making yeast are Trichoderma viride, Aspergillus niger, Hansenula sp, Candida sp. This study aims to determine the process of making a starter using rice flour and knowing the number of microbes that grow on pottato dextrose agar (PDA) media with concentrations of 0%, 4%, 8%, and 12%. This study used acompletely randomized design with 6 replications with 4 concentrations. Data analysis used ANOVA SPSS 17.1 program. The result of the analysis shows that the difference (P <0.05) is smaller than 0.05 (0.0152). From the result of the study, the starter of fungal microorganisms mixed with rice flour and PDA for the highest fungal growth was found at concentrations of 4%, 8% and 12%, namely 5,70, 6,14 and 6,13 with the addition of PDA media while at a concentration of 0% without PDA got 5,40 results.

ABSTRAK
Tepung beras merupakan bahan baku yang sering digunakan untuk pembuatan starter. Tepung beras memiliki kadar pati 90% dari berat kering. Starter merupakan kumpulan biakakn jamur yang siap diinokulasi untuk fermnetasi. Jamur yang biasa digunakan dalam pembuatan starter yaitu Trichoderma viride, Aspergillus niger, Hansenula sp, Candida sp. penelitian ini bertujuan untuk mengetahui proses pembuatan starter dengan menggunakan tepung beras dan mengetahui jumlah mikroba yang tumbuh pada media PDA dengan konsentrasi 0%, 4%, 8%, dan 12%. Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan 6 kali ulangan dengan 4 konsentrasi. Analisis data menggunakan ANOVA program SPSS 17.1. Hasil analisa didapatkan perbedaan nyata karena nilai P > 0,05 yaiitu (0,0152). Dari hasil penelitian starter mikroorganisme jamur yang dicampur dengan tepung beras serta PDA untuk pertumbuhan jamur tertinggi terdapat pada konsentrasi 4 %, 8% dan 12% yaitu 5,70, 6,14 dan 6,13 dengan penambahan media PDA sedangkan pada konsentrasi 0% (tanpa PDA) didaptkan hasil 5,40.
