Abstract

Lactate Dihydrogenase (LDH) enzyme levels can detect physiological changes in the body. LDH has 5 isoenzymes (LDH1, LDH2, LDH3, LDH4, LDH5) that is almost all the cells metabolism. LDH is one of an enzyme which is found in the blood serum in the heart. On specification of the heart there are LDH1 and LDH2. Heart muscle damage can be detected with an indicator of increased LDH levels. The aim of this research is to know the toxic effect of Metabolic Extract Scurrula atropurpurea (Bl.) Dans (MESA) on female wistar rats (Rattus norvegicus) subcronic for 28 days. The design of this research is True Experimental Design with Completely Randomized Design (RAL). MESA described 5 times a week for 28 days to tailed rat wistar female and divide into 4 treatment, 1treatment as control and 3 treatment as those who are doses different MESA the 250 mg/KgBW, 500 mg/KgBW and 1000 mg/KgBW. The rat in acclimatization for approximately 7 days before given the treatment. After 28 days, the animal tried in surgery and taken blood serum to test the levels of LDH. The results of LDH levels of the treatment were compared with the control. The results showed that the serum rat LDH concentration increased at 1000 mg/KgBW dose compared to control, but after the ANOVA test showed that the treatment rats were not significantly different from the rat control, meaning that MESA had no effect on the serum rat LDH females and no toxic effects are generated so that MESA is safe.

ABSTRAK


Kadar enzim Laktat Dihidrogenase (LDH) dapat mendeteksi perubahan fisiologis dalam tubuh. LDH memiliki 5 isoenzim yakni (LDH1, LDH2, LDH3, LDH4, LDH5) yang terdapat hampir semua sel yang bermetabolisme. LDH merupakan salah satu enzim yang terdapat pada serum darah yang berada dijantung. Pada spesifikasi jantung terdapat LDH1 dan LDH2. Kerusakan otot jantung dapat diketahui dengan indikator meningkatnya kadar LDH. Tujuan Penelitian ini untuk mengetahui efek toksik Ekstrak Metanolik Scurrula atropurpurea (Bl.) Dans  (EMSA) pada tikus wistar betina (Rattus norvegicus) secara subkronik selama 28 hari. Desain penelitian ini adalah True Experimental Design dengan Rancangan Acak Lengkap (RAL). EMSA dipaparkan 5 kali dalam seminggu selama 28 hari kepada 20 ekor tikus wistar betina dan dibagi menjadi 4 perlakuan, 1 perlakuan sebagai kontrol dan 3 perlakuan sebagai perlakuan yang diberi dosis EMSA yang berbeda yaitu 250 mg/KgBB, 500 mg/KgBB dan 1000 mg/KgBB. Sebelum diberi perlakuan tikus diaklimatisasi selama kurang lebih 7 hari. Setelah 28 hari, hewan coba dibedah dan diambil serum darahnya untuk dilakukan uji kadar LDH. Hasil kadar LDH perlakuan dibandingkan dengan kontrol. Hasil penelitian menunjukkan bahwa kadar LDH serum tikus mengalami peningkatan pada dosis 1000 mg/KgBB dibandingkan dengan kontrol, tetapi setelah dilakukan uji ANOVA menunjukkan bahwa tikus perlakuan tidak berbeda nyata dibandingkan dengan tikus kontrol. Hal ini berarti bahwa EMSA tidak berpengaruh terhadap kadar LDH serum tikus betina dan tidak ada efek toksik yang ditimbulkan sehingga EMSA bersifat aman.
