Abstract

Zebrafish are often chosen as an animal model because of their high physiological similarity to humans, their rapid life cycle, and their transparent embryos that facilitate direct visualization of organ development, making them an effective animal model for screening the effectiveness of natural agents and understanding disease mechanisms. This study aimed to evaluate the antihyperlipidemic effect of soursop leaf aqueous extract (Annona muricata L.) on hyperlipidemic animal model zebrafish (Danio rerio Hamilton, 1822). Hyperlipidemic conditions were obtained by induction using a 0.1% quail egg yolk suspension for 48 hours in zebrafish aged 5 days post fertilization. The concentration of extract was determined based on the MNLC (Maximum Non-Lethal Concentration) method. Based on the results of the MNLC test, the determined extract concentrations were 1/10 MNLC, 1/3 MNLC, and MNLC. The antihyperlipidemic effect was evaluated based on the measurement of total triglycerides and total cholesterol using spectrophotometric method and the results of Oil Red O (ORO) staining. The results of the spectrophotometric test showed that the most effective concentration of soursop leaf aqueous extract to reduce triglyceride and cholesterol levels in zebrafish larvae was the MNLC concentration. Soursop leaf aqueous extract with MNLC concentration significantly (p>0.05) reduced triglyceride and total cholesterol levels in zebrafish by 40.52 and 71.54%, respectively. Spectrophotometric measurements also positively correlated with the results of ORO staining, which showed a decrease in red color intensity. The results of this study indicate that soursop leaf aqueous extract has the potential to have antihyperlipidemic properties and can be studied in animal model zebrafish.
